Somatic embryogenesis and plantlet regeneration in red sandalwood (Pterocarpus santalinus)

Cotyledonary segments from the germinated immature zygotic embryos were used for somatic embryogenesis of red sandalwood (Pterocarpus santalinus). The explants established on Murashige and Skoog (MS) medium containing 5% sucrose combination 6-benzylaminopurine (BAP), 2,4-Dichlorophenoxyacetic acid (2,4-D), α-Naphthaleneacetic acid. All treatments responded to callus induction with a 36–97% frequency range. maximum embryogenic (69.44%) was obtained when 0.1 mg/l BAP + 2 2,4-D 4 combinations used. When treated individual growth regulators, (58.33%) produced by 2,4-D. completely ineffective individually. average number globular-staged ranged between 1 5 (irrespective treatments). cotyledonary-staged (2.85) treatment plantlets developed (1.30) transferred MS basal medium. acclimatized showed 100% survival in greenhouse condition. embryonic cells have been histologically distinguished non-embryonic dense cytoplasm long suspensor. induction, maturation, germination challenging, suggesting need molecular approaches through proteomic expression mass production understanding evolution, structure, genetic organization plant species. NAA, individually or BAP, has significant effect potential cotyledon explant P. santalinus.
